Analysis of oligomeric amyloid beta structures with single molecule fluorescence spectroscopy and FREThe amyloid beta peptide originates from specific cleavage of the APP (amyloid beta precursor protein). It aggregates to linear fibrils and is the main component of the amyloid beta plaques found in the brain of Alzheimer patients. However, recent results indicated that amyloid beta oligomers may be the main factor for neurotoxicity and the degradation of the brain. We apply single molecule FRET (fluorescence resonance energy transfer) to study the process of early amyloid beta aggregation. This allows us to measure accurate inter and intramolecular distances between fluorescently labeled amyloid beta peptides with sub nanometer accuracy. Thus the measured distances reveal the structure of the amyloid beta particles and their conformational changes during the early process of aggregation. The aim of this project is to provide a thorough characterization of the toxic oligomeric amyloid beta particles required for further studies on structural changes of the particles in CSF and the presence of new drug candidates..
